Amino-dPEG®4-OH, product number 10249, also known as Amino-dPEG®4-alcohol, is a short, monodisperse PEGylation reagent. This non-immunogenic, single molecular weight polyethylene glycol (PEG) reagent has a discrete-length chain. Therefore, it is sold under the dPEG® tradename. By reacting the amine end of this product with free carboxylates, this extremely hydrophilic PEGylation reagent modifies biomolecules and surfaces with appropriate functional groups.

PEGylation with dPEG® Products

Typical PEGylation reagents are disperse polymers. Such polymers are complex mixtures of different chain lengths and molecular weights. Conjugates made with non-uniform polymers are challenging to analyze.

In contrast, Quanta BioDesign’s dPEG® products are single molecular weight products with discrete PEG chain lengths. Although our products are monodispersed, we do not purify PEG from a polymer mixture. Instead, we synthesize our products as single molecules from high purity starting materials. Consequently, our monodisperse PEGylation reagents are superior to other types of PEGylation reagents.

For What Applications is Amino-dPEG®4-OH Used?

This product has many possible uses. The primary amine on one end of the molecule reacts with carboxylates to form amide bonds. Also, it reacts with aldehydes or ketones to form Schiff bases that are reducible to secondary amines. The most popular use for this product is the passivation of carboxylate-functionalized surfaces. The terminal alcohol of the PEG chain is non-immunogenic and imparts hydrophilicity to the conjugate molecule or surface. Consequently, conjugate molecules are water-soluble. Moreover, when used to passivate surfaces, Aminod-dPEG®4-OH reduces or eliminates non-specific binding.

Amino-dPEG®4-OH appears several times in published scientific literature. Applications of this product include:

  • modification and passivation of nanoparticle surfaces;
  • development of SPR sensors;
  • development of a confocal imaging system; and,
  • development of an electric field sensor for DNA polymerase.
