Amino-dPEG®24-acid, product number 10317, is a PEGylated amino acid. A primary amine group and a propionic acid group sit on opposite ends of a polyethylene glycol spacer. The highly hydrophilic, discrete PEG spacer (Ð = 1) imparts water solubility to conjugates that incorporate it. Moreover, Amino-dPEG®24-acid benefitted a variety of applications. These applications include peptide synthesis, surface modification, oligonucleotide sequencing, and small molecule modification.

Amino-dPEG®24-acid offers several benefits over comparable amino acids used as spacers. Among the product’s advantages, the following are notable:

  • Uniform PEG chain length and molecular weight;
  • Easily characterized, highly pure product;
  • Improved water solubility of conjugate molecules compared to the non-PEGylated product;
  • Increased hydrodynamic volume of conjugate molecules compared to the non-PEGylated product;
  • Enhanced flexibility of molecules that incorporate PN10317 into the backbone structure (g., a peptide-PEG construct);
  • Non-antigenic/non-immunogenic;

Both ends of the molecule are available for conjugation. Conjugating only one end of PN10317 to a target molecule may modify the overall charge of the conjugate. Many different types of coupling reactions can be used. However, avoid using a single-step EDC coupling, as this will polymerize the dPEG® product.

Several scientific publications and patents report the use of Amino-dPEG®24-acid in modifying diverse compounds. Among the various applications using this product are aptamers, quantum dots, therapeutic peptides, and DNA polymerase for nucleotide sequencing.

In addition to a dPEG®24 spacer, Quanta BioDesign offers related products containing longer or shorter spacers. These products include dPEG®4, dPEG®6, dPEG®8, dPEG®12, and dPEG®36 spacers.
