Amino-dPEG®36-acid, product number 10907, is a PEGylated amino acid. A primary amine group and a propionic acid group sit on opposite ends of a polyethylene glycol spacer. The highly hydrophilic, discrete PEG spacer (Ð = 1) imparts water solubility to conjugates that incorporate it. Moreover, Amino-dPEG®36-acid benefitted many, various applications. These applications include peptide synthesis, surface modification, oligonucleotide sequencing, and small molecule modification.

Amino-dPEG®36-acid offers several benefits over comparable amino acids used as spacers. Among the product’s advantages, the following are notable:

  • Uniform PEG chain length and molecular weight;
  • Easily characterized, highly pure product;
  • More water solubility of conjugate molecules compared to the non-PEGylated product;
  • Increased hydrodynamic volume of conjugate molecules compared to the non-PEGylated product;
  • Greatly enhanced flexibility of molecules that incorporate PN10907 into the backbone structure (g., a peptide-PEG construct);
  • Non-antigenic/non-immunogenic;

Both ends of the molecule are available for conjugation. Conjugating only one end of the molecule may modify the overall charge of the conjugate. Moreover, because either end of the molecule can be conjugated, carefully selected reaction conditions permit control of the conjugation, resulting in a well-defined product. By contrast, the use of comparable polymer PEG products will result in an intractable mixture of products. Many different types of coupling reactions can be used. However, users of this product should avoid employing a single-step EDC coupling, as this will polymerize the dPEG® product.

This product is useful for modifying the properties of various types of surfaces and biomolecules, peptides, proteins, dendrimers, aptamers. Notably, in a 2012 publication, researchers at Columbia University, Virginia Commonwealth University, and the National Institute of Standards (NIST) in Gaithersburg, Maryland, developed an inexpensive method for single-molecule DNA sequencing that incorporated Amino-dPEG®36-acid.

In addition to a dPEG®36 spacer, Quanta BioDesign offers related products containing different spacer lengths. These products include dPEG®4, dPEG®6, dPEG®8, dPEG®12, and dPEG®24 spacers.
