Amino-dPEG®24-amido-dPEG®24-acid, product number 11305, is a PEGylated amino acid that is useful for many, varied applications. Applications that may be improved by this product include peptide synthesis, surface modification, oligonucleotide sequencing, and small molecule modification.

The molecule contains a primary amine and a propionic acid group on opposite ends of a single molecular weight polyethylene glycol (PEG) spacer with a discrete chain length. The discrete PEG (dPEG®) spacer consists of two dPEG®24 chains joined via an amide bond in the middle of the chain. The total length of the dPEG® spacer is 152 atoms, which is a distance 178.9 Å. The highly hydrophilic, discrete PEG spacer (Ð = 1) imparts water solubility to conjugates that incorporate it. It is also soluble in many organic solvents.

Product number 11305 offers several benefits over comparable amino acids used as spacers. Among the product’s advantages, the following are significant:

  • Uniform PEG chain length and molecular weight;
  • Easily characterized, highly pure product;
  • Conjugate molecules have improved water solubility compared to the non-PEGylated product;
  • Increased hydrodynamic volume of conjugate molecules compared to the non-PEGylated product;
  • Greatly enhanced flexibility of molecules that incorporate PN11305 into the backbone structure (g., a peptide-PEG construct);
  • Non-antigenic/non-immunogenic;

Both ends of the molecule are available for conjugation. Conjugation of only one end of the dPEG® spacer may modify the overall charge of the molecule. Because either end of the molecule can be conjugated, carefully selected reaction conditions permit control of the conjugation, resulting in a well-defined product. By contrast, the use of comparable polymer PEG products will result in an intractable mixture of products. Many different types of coupling reactions are useful for conjugation. However, users of this product should avoid employing a single-step EDC coupling, as this will polymerize the dPEG® product.
