MAL-dPEG®24-Tris (-dPEG®24-Tris (m-dPEG®24)3)3, product number 11487, is a sulfhydryl-reactive, multibranched, monodispersed PEG product. The product is built around tris. One arm terminates in a maleimide group that reacts selectively with free thiols in a click chemistry type of reaction. Nine branches terminate in methyl groups. The amphiphilic PEG imparts tremendous water solubility to conjugate molecules and expands their hydrodynamic volume sufficiently to eliminate the renal clearance of the conjugates.

What is dPEG®?

The dPEG® tradename describes Quanta BioDesign’s patented brand of monodispersed PEGylation reagents. Unlike traditional PEGylation reagents, which are dispersed polymers, Quanta BioDesign’s dPEG® products are high-purity, single molecular weight PEG products with discrete chain lengths. PEGylation with MAL-dPEG®24-Tris(-dPEG®24-Tris(m-dPEG®24)3)3

MAL-dPEG®24-Tris(-dPEG®24-Tris(m-dPEG®24)3)3, product number 11487, is designed to modulate biodistribution in protein-PEG conjugates such as PEGylated antibody fragments and diabodies. The high molecular weight branched dPEG® compound imparts hydrophilicity to the conjugate and substantially increases the hydrodynamic volume. The increased hydrodynamic volume eliminates renal clearance, which can be a problem with small protein fragments. The dPEG® product also reduces the proteolysis of protein conjugates.

The maleimido group on one branch of the product reacts rapidly and specifically with sulfhydryl groups at pH < 7.5. Because cysteine residues are much less common in proteins than amino acid residues with free amine side chains (e.g., lysine), conjugation of MAL-dPEG®24-Tris (-dPEG®24-Tris (m-dPEG®24)3)3 can be directed to a few or one specific site on a protein. The terminal methyl groups on each of the remaining nine dPEG® branches of MAL-dPEG®24-Tris(-dPEG®24-Tris(m-dPEG®24)3)3 provide non-reactive termini.
