These novel products were designed for peptide synthesis applications and consist of Fmoc-protected lysine functionalized on the side chain with a biotin-dPEG® linker of either 4 or 12 PEG units length. This amino acid derivative can be incorporated anywhere it is needed in a growing peptide chain. The biotin-dPEG® side chain binds tightly to avidin or streptavidin and less strongly to other biotin-binding molecules. This design allows a peptide to function as a detection agent for targeted therapeutic or diagnostic applications. The Fmoc-protected lysine works in peptide synthesis as normal, and the Fmoc protecting group removes readily with piperidine. Using these products reduces the number of reaction steps and the amounts of reagents and solvents required for successful synthesis because no additional side-chain functionalization steps are required to incorporate biotin into the peptide chain.
